CELLULAR%20METABOLISM%20OF%20BREVETOXIN%20%28PBTX-2%29%20IN%20IMMUNE%20CELL%20LINES

The%20toxic%20dinoflagellate%2C%20Karenia%20brevis%2C%20produces%20brevetoxins%2C%20potent%20marine%20toxins%20which%20exert%20many%20harmful%20effects%20on%20sealife%20and%20cause%20severe%20respiratory%20symptoms%20in%20humans.%20Existing%20data%20suggest%20brevetoxins%20may%20affect%20immune%20system%20components.%20Preliminary%20data%20using%20a%20T%20lymphocyte%20cell%20line%20%28Jurkat%29%20and%20a%20monocyte%20cell%20line%20%28U-937%29%20as%20immune%20cell%20models%2C%20suggest%20immune%20cells%20may%20metabolize%20PbTx-2%20through%20conjugative%20metabolic%20pathways.%20Jurkat%20cells%20exposed%20to%2010%20%C2%B5g/ml%20PbTx-2%20for%202%20h%20demonstrated%20an%20approximately%2040%25%20decrease%20in%20intracellular%20glutathione%2C%20as%20measured%20using%20a%20colorimetric%20microplate%20assay.%20Using%20LC/MS%2C%20formation%20of%20a%20cysteine-PbTx-2%20conjugate%20%28m/z%201018%29%20and%20a%20putative%20glutathione-PbTx-2%20conjugate%20%28m/z%201204%29%20was%20demonstrated%20in%20U-937%20cells%20cultured%20with%2050%20uM%20PbTx-2.%20Over%20an%20incubation%20period%20of%203%20h%2C%20a%20shift%20in%20metabolites%20occurred%2C%20with%20the%20potential%20GSH-PbTx-2%20conjugate%20present%20in%20detectable%20levels%20as%20early%20as%201%20h%20and%20increasing%20in%20concentration%20after%202%20h.%20The%20cysteine-PbTx-2%20conjugate%20was%20not%20detectable%20until%20after%202%20h%20of%20incubation.%20Depletion%20of%20intracellular%20glutathione%20and%20formation%20of%20brevetoxin%20metabolites%20is%20suggestive%20of%20active%20cellular%20metabolism%20of%20PbTx-2%20by%20immune%20cells%20in%20vitro.%20
